Purity >98% by SDS-PAGE and HPLC analyses.
Biological Activity Fully biologically active when compared to standard. The ED50 as determined by its ability to inhibit sRANK Ligand induced nuclear factor kappa B(NFkappaB) in RAW 264.7 cells is less than 50ng/ml, corresponding to a specific activity of >2.0×104IU/mg in the presence of 15ng/ml of recombinant sRANK Ligand.

Formulation Lyophilized from a 0.2μm filtered concentrated solution in 20mM Tris-HCl, pH8.0, 150mM NaCl.

Reconstitution We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Reconstitute in sterile distilled water or aqueous buffer containing 0.1% BSA to a concentration of 0.1-1.0mg/ml. Stock solutions should be apportioned into working aliquots and stored at ≤-20℃. Further dilutions should be made in appropriate buffered solutions.

Background RANK Ligand and RANK Receptor are members of the TNF superfamily that play an important role in the regulation of specific immunity and bone turnover. RANK Receptor was originally identified as a dendritic-cell-membrane protein, which by interacting with RANKL augments the ability of dendritic cells to stimulate naïve T cell proliferation and to promote the survival of RANK + T cells. RANK is also expressed in a variety of tissues including skeletal muscle, thymus, liver, colon, small intestine and adrenal gland. The RANK/RANKL interaction is important in the regulation of osteoclastogenesis and in dendritic-cellmediated T cell immune responses. Impairments in RANK signaling have been implicated in the induction of expansile osteolysis and Paget disease of bone (PDB2). Recombinant human sRANK receptor is a 19.1kDa polypeptide containing the TNFR homologous cysteine rich portion of the extracellular domain of RANK receptor (174 amino acid residues).

保存条件:
-20℃或更低温度保存,至少一年有效。由于蛋白的每次冻融均会引起部分失活,所以首次配制成相应浓度的储存液后(请根据产品简介中Reconstitution一栏的信息配制储存液),须分装后-20℃或更低温度冻存,以避免反复冻融。
注意事项:
由于有些塑料管壁对某些蛋白有较强的吸附作用,溶液中的蛋白很容易粘附在管壁上,并且粘附后的蛋白很难与管壁分离。而载体蛋白(Carrier protein,如0.1% BSA等)的主要作用是预先封闭塑料管壁上的蛋白结合位点,使细胞因子或重组蛋白不会粘附于管壁。所以一定要使用产品简介中Reconstitution一栏的信息配制储存液。

使用说明:
1. 收到产品后请立即按照说明书推荐的条件保存。
2. 将本产品和自行准备的产品简介中Reconstitution一栏推荐的配制储存液平衡至室温。
3. 除非特别注明,碧云天相关产品均为冻干粉,由于微量的蛋白在冻干过程中沉积在管内,形成很薄或不可见的蛋白层,所以在打开盖子前,我们建议在离心机中约8,000-12,000×g离心10-30秒,使附着在盖子或容器壁上的蛋白聚集于容器底部。
4. 加入适量推荐的配制储存液,达到Reconstitution一栏中推荐的浓度。请勿用vortex剧烈振荡或移液器剧烈吹打,以免蛋白变性而失活。
5. 在室温下溶解15-30分钟。
6. 如果看不到蛋白粉末,并担心蛋白仍然粘附在盖子或容器壁上,可以吸取液体轻轻润洗盖子和容器壁以促进溶解。对于溶解的体积相对比较大的情况,可使用BeyoVortex™基础型旋转混匀仪(E6800)或其它适当摇床或手动轻轻上下颠倒以促进溶解。
7. 如需分装,每管体积通常不小于10μl,并按说明书中推荐的储存液保存条件进行保存。
8. 具体的最佳工作浓度请自行参考相关文献,或者根据实验目的,以及特定细胞和动物,通过实验进行摸索和优化。
